I’m the Chairman and CEO of Advaxis, a clinical-stage biotechnology company that is enthusiastic about what we do.</p>
<p>I want to share with you what makes our company unique among next generation immunotherapies- our technology and our people. This blog is intended to provide insight into recent developments in biotechnology and where we fit in the expanding biopharmaceutical landscape.</p>
<p>Technology can be as reflective of cultural change as fashion or politics. Immunotherapy’s focus on helping the body heal itself appeals to our current appreciation of the organic, the holistic, and our higher standards for quality of life during treatment.</p>
<p>Chemotherapy, radiation, and surgery can be effective against cancer, but can come at a great cost to the patient. The American Cancer Society cautions that chemotherapy and radiation can have long-lasting side effects over months or years.<a title="" href="#_ftn1">[1]</a> The next generation of cancer treatments is intended to be as effective while more narrowly targeting the rapidly dividing cells that characterize cancer.</p>
<p>As the 17th century Spanish writer Baltasar Gracián said, “You may be obliged to wage war, but not to use poison arrows.”<a title="" href="#_ftn2">[2]</a> Refocusing immune defenses to fight tumor cells has the potential to be highly effective, specific in its attack, and with less collateral cellular damage. It’s ironic that three immunologists recently awarded the Nobel Prize would be given an award named after the man who invented dynamite. Immunotherapies are designed to be the opposite of a general blasting away at cancer, as we have had to do to date.</p>
<p>The first generation of immunotherapies has been focused on harnessing the body’s defense mechanisms. The goal of the next generation is to develop immunotherapies that further diminish side effects. “Do no harm” is an oath as old as the medical profession, but the next generation of immunotherapies is truly taking that notion to heart.</p>

This intellectual property resulted from research conducted in Dr. Yvonne Paterson’s laboratory that demonstrated ISG15 was an effective immunological target for the treatment of breast cancer in animal models.</p>
<p>&#8220;Targeting ISG15 may represent a therapeutic approach suitable for a population of patients with advanced breast cancer for whom few approved treatments currently exist. The research conducted by Dr. Paterson and colleagues, as well as research conducted at other institutions, suggests that this antigen may have utility in treating other tumor types as well. 			<content:encoded><![CDATA[http://www.advaxis.com/uncategorized/advaxis-receives-additional-financing/Advaxis Receives Additional Financing<div><a href="" title=""></a></div><p><strong>Princeton, NJ – October 28, 2011 – </strong><a href="http://www.advaxis.com/"><strong>Advaxis, Inc</strong></a>.<strong>, (OTCBB: ADXS)</strong>, a leader in developing the next generation of immunotherapies for cancer and infectious diseases, has entered into a definitive Convertible Notes Purchase Agreement (Agreement) for the purchase of the aggregate principal value of approximately $2.3 million of Convertible Promissory Notes (Notes) for an aggregate purchase price of approximately $2.0 million. Investors include certain accredited investors as well as the CEO and the CFO of the Company.  The Notes purchased in the offering shall be paid for in cash or, with respect to Notes acquired by Advaxis Chairman/CEO Thomas A. Moore, in exchange for the cancellation of $400,000 of outstanding indebtedness owed by the Company.  The closing of the sale of the Notes, under the Agreement, is anticipated to occur on or about October 31, 2011, subject to customary closing conditions.</p>
<p>The Notes have an original issue discount of 15% and mature on the one year anniversary of the issue date. Under the terms specified therein, the Notes are convertible, in whole or in part, into common stock at a conversion price of $.15. However, except as otherwise provided in the Notes, only 85% of the initial principal amount of each Note is convertible at any time after issuance and the remainder is convertible at maturity.</p>
<p>In connection with the Agreement, the Company issued a warrant to each investor to acquire up to such number of shares of Common Stock equal to 50% of such number of Conversion Shares, issuable upon exercise of the Note issued to such Investor as of the Closing Date with an exercise price of $0.15. The warrants will expire in three years. Pursuant to a Registration Rights Agreement between the Company and the investors, the Company is required to file a resale registration statement within 30 days that covers the resale of the underlying shares of both the Notes and shares issuable upon exercise of the warrants.</p>
<p>“This financing will provide additional operating funds between now and the announcement of interim results for our Phase 2 clinical trial on cervical cancer currently underway in India. I am particularly pleased that many of those supporting this financing have invested in the company previously,” commented Advaxis Chairman/CEO Thomas A. Moore. “This small group of investors also includes the company’s chief financial officer and myself.”</p>

			<content:encoded><![CDATA[http://www.advaxis.com/uncategorized/advaxis-announces-notice-of-allowance-of-us-patent-for-adxs-hpv-immunotherapy/Advaxis Announces Notice of Allowance of US Patent for ADXS-HPV Immunotherapy<div><a href="" title=""></a></div><p><strong>Princeton, NJ – October 18, 2011 – </strong><a href="http://www.advaxis.com/"><strong>Advaxis, Inc</strong></a>.<strong>, (OTCBB: ADXS)</strong>, a leader in developing the next generation of immunotherapies for cancer and infectious diseases, today announced that the United States Patent and Trademark Office has issued a Notice of Allowance for a US Patent titled “Compositions and Methods for Treatment of Cervical Cancer.” This patent specifically protects the use of the Advaxis HPV construct for the treatment of late stage cervical cancer, an unmet medical need for which this agent is currently being evaluated in 2 Phase II clinical trials, two of which are in recurrent metastatic cervical cancer, one in India and one in collaboration with the Gynecologic Oncology Group of the National Cancer Institute in the US.</p>
<p>Once issued, this patent will provide a new 17+ year patent cycle for Advaxis’ lead compound.</p>
<p>This patent establishes ADXS-HPV as a method of treatment for late stage cervical cancer in human patients using a recombinant <em>Listeria</em> strain comprised of a fusion of HPV E7 antigen and an N-terminal fragment of an LLO protein.</p>
<p>This patent also protects ADXS-HPV in various physical formulations, including a dried formulation that is stable at room temperature.</p>
<p>The subject matter of this patent is based on the work of Dr. Yvonne Paterson and Dr. John Rothman and is assigned to both the University of Pennsylvania and Advaxis. Together, Advaxis and the University of Pennsylvania hold over 70 approved patents and pending patent applications for composition of matter and methods of use.</p>

<p>A pre-IND meeting is conducted to develop mutual understanding and agreement between the FDA and the Company regarding the content needed to assess manufacturing, toxicology, pre-clinical and rationale to support a first in humans clinical trial. Once an IND is submitted, the FDA has 30 days to comment before the Company can begin Phase 1 trials. The Company plans to file the IND application early in 2012 to initiate a Phase 1/2 safety study in humans.</p>

<p>HER2 (human epidermal growth factor receptor 2) is a gene which is over expressed in a percentage of certain types of cancers such as breast, stomach, bladder, pancreatic, brain, and ovarian. The American Cancer Society estimates that in 2011 in the US there will be 230,480 diagnoses of invasive breast cancer, 21,520 new cases of stomach cancer, 69,250 new cases of bladder cancer, 44,030 new cases of pancreatic cancer, 22,340 new cases of brain/spinal cord cancer, and 21,900 new cases of ovarian cancer.</p>
